You should have;

Solid experience of User Interface development
Expert knowledge of XHTML and CSS
Expert knowledge of accessibility standards
Knowledge of Javascript, JQuery and AJAX
Expert knowledge of browser capabilities
Knowledge of either Java J2EE or ASP.Net
Agile experience
Adobe creative suite ideas as is IDE experience (i.e., Visual Studio)
